中文摘要:目的 研究酸枣仁对失眠模型大鼠下丘脑-垂体-肾上腺(HPA)轴功能的影响。方法 连续3 d采用腹腔注射对氯苯丙氨酸(PCPA)复制失眠大鼠模型,模型复制同期给药。给药组分为酸枣仁乙醇提物高剂量组(20 g·kg–1·d–1)和低剂量组(5 g·kg–1·d–1)。给药10 d后,观察各组大鼠行为;苏木精-伊红(HE)染色评价HPA轴相关组织的病理改变;酶联免疫吸附测定法(ELISA)检测血清及HPA轴组织中的促肾上皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)水平,以及血清白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)含量。结果 PCPA致失眠大鼠模型复制成功,给药酸枣仁提取物可改善失眠引起的大鼠行为异常,缓解模型大鼠HPA轴组织病理改变,抑制模型大鼠血清及HPA轴组织中CRH、ACTH、CORT水平升高,促进模型大鼠血清IL-1β、TNF-α表达量的回调。结论 酸枣仁可通过调节HPA轴功能达到干预失眠症发生与发展的作用。

Effect of Ziziphi Spinosae Semen on HPA Axis Function in Rats with Insomnia
Abstract:Objective To study the effect of Ziziphi Spinosae Semen (ZSS) on the function of the hypothalamic-pituitary-adrenal (HPA) axis in rats with insomnia.Methods The rat model of insomnia was established by intraperitoneal injection of para-chlorophenylalanine (PCPA) for 3 days. At the same time, the rats in each group were given the ZSS extract by gavage for 10 days. The administration groups were divided into a high-dose ZSS extract (20 g·kg–1·d–1) group and a low-dose ZSS extract (5 g·kg–1·d–1) group. The behavior of rats in each group was observed after 10 days of administration. The pathological changes in HPA axis-related tissues were evaluated by hematoxylin-eosin (HE) staining. The levels of corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH), and corticosterone (CORT) in serum and HPA axis-related tissues were determined by enzyme-linked immunosorbent assay (ELISA). The content of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in serum were also determined by ELISA.Results The rat model of insomnia induced by PCPA was successfully replicated. Administration of ZSS extract improved the behavioral abnormalities of rats caused by insomnia, alleviated the pathological changes in the HPA axis, inhibited the increase in the levels of CRH, ACTH, and CORT in serum and HPA axis, and promoted the expression callback of IL-1β and TNF-α in serum of rats with insomnia.Conclusion ZSS extract influence the occurrence and development of insomnia by inhibiting the function of the HPA axis.
