中文摘要:目的 研究保健保本草菁萃作为保健食品使用的安全性及对小鼠免疫功能的影响。方法 采用小鼠急性经口毒性实验、Ames实验和大鼠30 d喂养实验对受试物进行毒理学研究和评价;采用小鼠脾淋巴细胞转化实验、迟发型变态反应实验、抗体生成细胞检测实验、小鼠腹腔巨噬细胞吞噬鸡红细胞实验和乳酸脱氢酶活性测定实验,从细胞免疫功能、体液免疫功能、单核-巨噬细胞功能及自然杀伤(NK)细胞活性4个方面综合评价受试物对小鼠免疫功能的影响。结果 受试物急性经口剂量在38.83 g·kg–1·d–1时对雌、雄小鼠没有毒性;加与不加大鼠肝匀浆S9混合液时,Ames实验结果均为阴性,未见致突变作用;30 d喂养实验中,大鼠生长发育良好,血液学、血液生化学、组织学检查指标与主要脏器系数与对照组相比差异均无统计学意义;受试物能促进刀豆蛋白A(ConA)诱导的淋巴细胞增殖,具有调节细胞免疫功能;能提高NK细胞活性,具有调节非特异性免疫功能。结论 保健保本草菁萃具有较好的安全性,可以调节小鼠免疫功能,因而可作为一种功能性食品补充剂。 |

Toxicological Safety Evaluation of Inpolysac Authentic Herbal Mix and Its Effect on Immune Function in Mice |

Abstract:Objective To study the toxicological safety of Inpolysac Authentic Herbal Mix as a health food and its effect on immune function in mice.Methods The toxicological safety of the tested substance was assessed by acute oral toxicity test in mice, Ames test, and 30-day feeding test in rats. Mouse spleen lymphocyte transformation test, delayed hypersensitivity test, antibody-producing cell test, mouse peritoneal macrophage phagocytosis of chicken red blood cells test, and lactate dehydrogenase test were employed to evaluate the effect of the tested substance on mouse immune function from four aspects: cellular immune function, humoral immune function, monocyte-macrophage function, and NK cell activity.Results In the acute oral toxicity test, when the dose was 38.83 g·kg–1·d–1, the tested substance had no toxic reactions in male and female mice. The results of Ames test were negative with or without S9 and no mutagenicity was observed. In the 30-day feeding test, the experimental animals grew and developed well, and there was no significant difference in hematology indexes, blood biochemistry indexes, main organ coefficients, and histological examination between the experimental groups and the control group. The tested substance can promote the proliferation of lymphocytes induced by ConA and increase the activity of NK cells and it had the functions of regulating cellular immunity and non-specific immunity.Conclusion Inpolysac Authentic Herbal Mix is safe. It can regulate the immune function of mice, which can be used as a kind of functional food supplement. |
