中文摘要:目的 建立不同产地白术炮制前后新绿原酸和绿原酸的含量测定方法,比较炮制前后含量变化。方法 采用高效液相色谱法,测定15批不同产地的白术药材和饮片中新绿原酸和绿原酸的含量,并通过GraphPad Prism 8软件对各产地白术炮制前后新绿原酸和绿原酸含量进行单因素方差分析。结果 不同产地白术新绿原酸和绿原酸含量存在差异,其中河北产的白术绿原酸质量分数为0.216 3%,显著高于浙江和安徽产;炮制后新绿原酸和绿原酸含量均有所下降,转移率最高的是浙江产白术,新绿原酸和绿原酸平均转移率分别为55.68%、55.05%。结论 建立的含量测定方法简单、准确、稳定,适用于白术及其炮制品中的新绿原酸和绿原酸含量测定,并初步揭示了白术炮制前后新绿原酸和绿原酸含量的变化情况。

The Content Change of Neochlorogenic Acid and Chlorogenic Acid in Atractylodes macrocephala Koidz. from Different Producing Areas and Its Processed Products
Abstract:Objective To establish a method for the determination of neochlorogenic acid and chlorogenic acid in Atractylodes macrocephala Koidz. from different producing areas and its processed products, and compare the content changes of neochlorogenic acid and chlorogenic acid before and after processing.Methods High performance liquid chromatography was conducted, the contents of neochlorogenic acid and chlorogenic acid in 15 batches of A. macrocephala and decoction pieces from different origins were determined, and one-way analysis of variance was performed on the contents of neochlorogenic acid and chlorogenic acid in A. macrocephala before and after processing by GraphPad Prism 8.Results There were differences in the contents of neochlorogenic acid and chlorogenic acid in A. macrocephala from different producing areas. The content of chlorogenic acid in A. macrocephala from Hebei was 0.216 3%, which was significantly higher than that from Zhejiang and Anhui. After processing, the contents of neochlorogenic acid and chlorogenic acid decreased, and the transfer rate was the highest in A. macrocephala from Zhejiang, with the average transfer rate of neochlorogenic acid and chlorogenic acid reaching 55.68% and 55.05%, respectively.Conclusion The content determination method established in this study was simple, accurate and stable, and was suitable for the determination of neochlorogenic acid and chlorogenic acid in A. macrocephala. This study preliminarily revealed the content changes of neochlorogenic acid and chlorogenic acid before and after processing.
