中文摘要:目的 探索唐古特大黄外观性状与内在成分含量之间的关联性,为唐古特大黄的“辨状论质”提供理论依据。方法 采用质构仪、分光测色仪和电子舌、高效液相色谱法测定唐古特大黄根和根状茎的4项外观性状指标,以及15个药效成分指标,并将外观性状指标与药效成分含量进行相关性分析和聚类分析。结果 外观性状与药效成分含量相关性趋势在根和根状茎中基本一致。整体来看,根和根状茎的粗度和硬度越大、颜色偏橙色且暗[明度值(L*)小,红色值(a*)、黄色值(b*)大]、苦味越小,药材中的没食子酸、番泻苷A、番泻苷B、大黄酸、大黄素及总蒽醌含量越高。不同的是,大黄酚、大黄素甲醚均在颜色偏橙色(a*b*大)的根中含量较高,大黄酚在颜色偏黄色(a*小、b*大)的根状茎中含量较高,大黄素甲醚在颜色偏褐色(a*b*小)的根状茎中有较高含量。结论 唐古特大黄外观性状指标(粗度、硬度、颜色、苦味大小)可作为“辨状论质”判断大黄质量的依据,且传统认为的根茎粗大、质坚实、颜色棕红或黄色为佳可以在根和根状茎的内在质量中得到体现,但传统认为的“味苦为佳”未在本研究中得到证实。

Correlation Between Appearance Traits and Internal Quality of Rheum tanguticum Maxim. ex Balf. Based on Theory of Quality Evaluation Through Morphological Identification
Abstract:Objective To explore the correlation between appearance traits and the content of intrinsic components of Rheum tanguticum Maxim. ex Balf., providing a theoretical basis for "quality evaluation through morphological identification".Methods Four appearance trait indicators and 15 medicinal component indicators of R. tanguticum roots and rhizomes were determined using texture analyzer, spectrophotometer, electronic tongue, and high-performance liquid chromatography (HPLC). Correlation analysis and cluster analysis were conducted between the appearance trait indicators and the content of medicinal components.Results The correlation trends between appearance traits and medicinal component content were generally consistent in both the roots and rhizomes. Overall, R. tanguticum with larger and harder roots and rhizomes, more orange and dark color (lower L*, higher a* and b*), and less bitter taste, had higher content of gallic acid, sennoside A, sennoside B, rhein, emodin, and total anthraquinone. The difference lay in the fact that the content of chrysophanol and physcion was higher in roots with a more orange color (higher a* and b*), while the content of chrysophanol was higher in rhizomes with a more yellow color (lower a*, higher b*), and the content of physcion was higher in rhizomes with a more brown color (lower a* and b*).Conclusion Appearance trait indicators of R. tanguticum (thickness, hardness, color, and bitterness) can serve as a basis for evaluating the quality of R. tanguticum. The traditional belief that larger, harder roots with brown-red or yellow color are preferable is reflected in the intrinsic quality of both the rhizomes and roots. However, the traditional preference for a bitter taste is not confirmed in this study.
