中文摘要:目的 建立基于2个引物组合的DNA指纹图谱技术,鉴别少棘蜈蚣药材及其混伪品。方法 收集蜈蚣属少棘蜈蚣及11个易混淆种共39批样品,使用通用鉴别引物12S-L1091/12S-H1478和16S-L3428/16S-H3667扩增并进行短串联重复序列(STR)分型,获得DNA指纹图谱。结果 少棘蜈蚣12S迁移峰为420.1 bp或424.7~426.8 bp,16S迁移峰为175.5~175.6 bp,均不同于其他物种。结论 基于荧光STR分型原理建立了一种简单快捷、可用于蜈蚣药材及其混伪样品的通用DNA指纹图谱鉴别,能够完成对少棘蜈蚣药材的鉴定,并且通过2个引物结合能够鉴别蜈蚣属其他物种。 |

Identification of Scolopendra subspinipes by DNA fingerprinting |

Abstract:Objective A DNA fingerprinting technique based on two primer combinations was established to identify Scolopendra subspinipes mutilans L.Koch and its adulterants.Methods A total of 39 batches of Scolopendra species were collected, including S. subspinipes and 11 easily confused species. DNA fingerprints were obtained using universal identification primers 12S-L1091/12S-H1478 and 16S-L3428/16S-H3667 as well as short tandem repeat (STR) typing.Results The DNA fingerprinting pattern for S. subspinipes at the 12S locus was either 420.1 bp or in the range of 424.7-426.8 bp, while at the 16S locus, it was in the range of 175.5-175.6 bp, distinguishing it from other species and enabling its identification.Conclusion A rapid and convenient method has been established based on STR typing for identifying Scolopendra species and their adulterants by DNA fingerprinting, thereby achieving the discrimination of S. subspinipes. Moreover, other Scolopendra species can be identified by the combination of these two primers. |
