中文摘要:目的 以云南道地药材滇黄精为研究对象,测定不同蒸晒条件下多糖和还原糖含量的变化,确定炮制工艺后,通过秀丽隐杆线虫(以下简称线虫)自然衰老和氧化应激模型,探究其对线虫寿命、体长、应激能力和抗氧化能力的影响。方法 通过苯酚-硫酸法和3,5-二硝基水杨酸法分别测定炮制过程中滇黄精多糖与还原糖含量的变化;建立线虫自然衰老和氧化应激等模型,观察不同给药浓度下秀丽隐杆线虫寿命、脂褐素含量、急性热应激能力、抗氧化应激能力等指标的改变。结果 滇黄精“二蒸二晒”后所含多糖和还原糖质量分数均最高,分别为4.792%和27.040%;相较于对照组,“二蒸二晒”组卵期8 d线虫脂褐素含量降低24.11%(P<0.01);选择“二蒸二晒”为滇黄精炮制工艺并提取多糖,结果显示质量浓度为0.05、1.00、20.00 mg·mL–1的滇黄精多糖分别使线虫寿命增长14.62%、51.90%(P<0.05)、12.02%(P<0.05);8 h急性热应激条件存活率分别提高17.63%、43.90%(P<0.01),降低9.01%(P<0.05);急性氧化应激条件存活率分别提高0.82%、42.54%(P<0.05)、33.78%(P<0.05)。结论 滇黄精“二蒸二晒”后多糖和还原糖含量最高,是较佳的炮制工艺;滇黄精多糖可延长线虫寿命,提高其抗应激能力。

Processing Technology and Anti-aging Activity of Polygonatum kingianum Based on Polysaccharide Content
Abstract:Objective To determine the changes of polysaccharides and reducing sugar in Polygonatum kingianum in Yunnan province under different steaming and sun-drying conditions, and to explore the effects of polysaccharides on the life span, body length, stress capacity and antioxidant capacity of Caenorhabditis elegans by constructing natural aging and oxidative stress models.Methods The changes of polysaccharides and reducing sugar in P. kingianum during processing were determined by phenol-sulfuric acid method and 3,5-dinitrosalicylic acid (DNS) assay, respectively. Natural aging and oxidative stress models of C. elegans were constructed to observe the changes of its life span, lipofuscin content, acute heat stress capacity and antioxidant capacity under different administration concentrations.Results The contents of polysaccharides (4.792%) and reducing sugar (27.040%) were the highest after two steaming and two sun-drying. Compared with that in control group, the lipofuscin content in C. elegans at 8 d of egg stage decreased by 24.11% (P<0.01) after two steaming and two sun-drying. The polysaccharides in P. kingianum at the concentrations of 0.05, 1.00 and 20.00 mg·mL–1 extracted by two steaming and two sun-drying increased the life span of C. elegans by 14.62%, 51.90% (P<0.05) and 12.02% (P<0.05), respectively; the survival rate under 8 h acute heat stress condition increased by 17.63%, 43.90% (P<0.01), and decreased by 9.01% (P<0.05), respectively; the survival rate under acute oxidative stress increased by 0.82%, 42.54% (P<0.05) and 33.78% (P<0.05), respectively.Conclusion The contents of polysaccharides and reducing sugar are the highest after two steaming and two sun-drying, which is a desirable processing technology. The polysaccharides can prolong the life of C. elegans and improve its anti-stress activity.
