中文摘要:目的 分析固本泄浊饮对慢性肾衰竭(CRF)大鼠肾功能生化指标、肾脏病理学变化及肠道菌群结构组成与功能上的影响,研究其治疗CRF的作用机制。方法 取SD大鼠随机分为6组:假手术(SHAM)组、模型(MOD)组、氯沙坦钾(LP,30 mg·kg–1·d–1)组、固本泄浊饮低、中、高剂量(GBXZYL、GBXZYM、GBXZYH,0.538、1.076、2.152 g·mL–1)组,造模成功后按分组给药8周。给药结束后,采集血液、肾组织和粪便进行生化检测、病理组织染色、16S rRNA测序分析。结果 在药效方面,各剂量固本泄浊饮均能有效调节相关生化指标,缓解肾组织炎症反应,尤其是GBXZYM组。在肠道菌群方面,GBXZYL、GBXZYM组可提升肾衰竭大鼠肠道菌群的多样性,各剂量组均能有效调节肠道菌群的组成和相对丰度,Parabacteroides和Blautia可能是固本泄浊饮调节肠道微生物的关键标志物种。在差异代谢通路方面,物质能量代谢和激素生物合成可能是固本泄浊饮对肠道菌群调节作用的潜在机制。结论 固本泄浊饮可通过调节肠道菌群结构、组成及差异成分改善5/6肾切除大鼠引起的CRF。

Effect of Guben Xiezhuo Decoction on Chronic Renal Failure in Rats by Regulating Gut Microbiota
Abstract:Objective To analyze the effect of Guben Xiezhuo Decoction on the renal biochemical indexes and pathology as well as the structure and function of gut microbiota of chronic renal failure (CRF) rats, and to reveal the mechanism in the treatment of CRF.Methods SD rats were randomly divided into sham operation group (SHAM group), model group (MOD group), losartan potassium group (LP group, 30 mg·kg–1·d–1), low-dose Guben Xiezhuo Decoction group (GBXZYL group, 0.538 g·mL–1), medium-dose Guben Xiezhuo Decoction group (GBXZYM group, 1.076 g·mL–1) and high-dose Guben Xiezhuo Decoction group (GBXZYH group, 2.152 g·mL–1). After successful modeling, the rats were administered for 8 weeks. The blood, kidney tissue and stool were collected for biochemical, pathological and 16S rRNA sequencing analysis after administration.Results In terms of efficacy, Guben Xiezhuo Decoction at different doses effectively regulated the relevant biochemical indexes and alleviated the inflammatory reaction of renal tissue, especially at medium dose. In gut microbiota, GBXZYL、GBXZYM groups enhanced the diversity of gut microbiota in CRF rats, and all dose groups effectively regulated the composition and relative abundance of gut microbiota. Paraacteroides and Blautia might be key species markers in Guben Xiezhuo Decoction regulating gut microbiota. Regarding differential metabolic pathway, material and energy metabolism as well as hormone biosynthesis might be the potential mechanism in the regulation of gut microbiota by Guben Xiezhuo Decoction.Conclusion Guben Xiezhuo Decoction can improve the 5/6 nephrectomy-induced CRF rats by regulating the structure, composition and differential components of gut microbiota.
