中文摘要:目的 建立济川煎HPLC指纹图谱,并进行化学模式识别评价不同产地饮片对济川煎质量的影响。方法 使用不同产地饮片制备济川煎样品,利用Caprisol C18-AQ 100A色谱柱(250 mm×4.6 mm,5 μm),乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,测定了15批济川煎的指纹图谱,结合相似度评价、聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)模式识别方法,对不同产地饮片制备的济川煎进行质量分析。结果 建立的指纹图谱方法符合方法学要求,15批济川煎HPLC指纹图谱有13个共有特征峰,指认了3个特征峰成分,其相似度均在0.9以上;通过CA发现,样品主要受枳壳产地影响,可分为两大类;PCA与CA结果基本一致;OPLS-DA筛选出了不同批次济川煎的2个差异性质量标志物,通过对照品指认,确定了8号为柚皮苷。结论 建立的指纹图谱方法稳定可靠,结合化学模式识别可将济川煎按枳壳产地不同分为2类,差异性质量标志物为柚皮苷,可用于济川煎的质量控制评价。 |

Establishment of HPLC Fingerprints and Chemical Pattern Recognition of Jichuan Decoction from Different Regions |

Abstract:Objective To establish HPLC fingerprints of Jichuan Decoction (JD) and perform chemical pattern recognition to evaluate the effect of different regions of medicinal materials on the quality of JD.Methods JD samples were prepared by using medicinal materials from different regions, and Caprisol C18-AQ 100A chromatographic column (250 mm×4.6 mmm, 5 μm) was used for gradient elution, with acetonitrile-0.1% phosphoric acid aqueous solution as mobile phase. Fingerprints of 15 batches of JD were established. Combined with similarity evaluation, cluster analysis (CA), principal component analysis (PCA), orthogonal partial least squares-discriminant analysis (OPLS-DA), and other pattern recognition methods, the quality of JD prepared by using medicinal materials from different regions was analyzed.Results The established fingerprint method met the methodological requirements. There were 13 common characteristic peaks in HPLC fingerprints of 15 batches of JD, and three characteristic peak components were identified. Their similarity was above 0.9. The CA showed that the samples could be divided into two categories according to the different origins of Citrus aurantium L. The results of PCA and CA were consistent. OPLS-DA was used to screen out two different markers of different batches of JD. Through the identification of the reference substance, it was determined that peak 8 was naringin.Conclusion The established fingerprint method is stable and reliable. Combined with chemical pattern recognition, JD could be divided into two categories according to the different origins of C. aurantium L. The different marker is naringin, and it can be used for the quality evaluation of JD. |
