中文摘要:目的:建立基于抗氧化谱效关系的肉苁蓉质量标志物(Q-Marker)研究方法,为其药效物质基础及质量控制研究提供依据。方法: 采用超高效液相色谱法(UPLC)建立10批肉苁蓉指纹图谱。利用过氧化氢(H2O2)建立细胞氧化损伤模型,以活性氧自由基半数抑制率(ROS IC50)、超氧化物歧化酶(Superoxide Dismutase,SOD)和过氧化氢酶(Catalase,CAT)含量为药效指标测定肉苁蓉抗氧化能力。通过主成分分析、Pearson相关性分析和偏最小二乘回归分析共同构建谱效关系,并对筛选出的活性成分进行药效验证。结果:建立了肉苁蓉的UPLC指纹图谱,共确定了17个共有峰。基于谱效关联分析筛选出9个成分,活性成分验证结果显示京尼平苷酸、8-表马钱子酸、松果菊苷、毛蕊花糖苷、异毛蕊花糖苷、管花苷A具有抗氧化活性且呈浓度依赖性。结论:基于谱效关系探讨了肉苁蓉的Q-Marker,为其药效物质基础及质量标准提升提供参考。

Study on Antioxidant Quality Markers of Cistanches Herba Based on Spectrum-effect Relationship
Abstract:Objective: To establish a quality marker research method of Cistanches Herba (CH) based on the spectrum-effect relationship of antioxidant, so as to provide a basis for the research on the pharmacodynamic material basis and quality control. Methods: UPLC was used to establish the fingerprints of 10 batches of CH. The oxidative damage model was established by hydrogen peroxide (H2O2). The antioxidant capacity of CH was determined by ROS IC50, SOD and CAT contents. Principal component analysis (PCA), Pearson correlation analysis and partial least squares regression (PLS) were used to establish the spectrum-effect relationship, and the efficacy of the selected active ingredients was verified. Results: The UPLC fingerprint of CH was established, and a total of 17 common peaks were identified. Nine components were screened out based on spectrum-efficacy correlation. The results of active ingredient verification showed that geniposidic acid, 8-epiloganic acid, echinacoside, acteoside, isoacteoside and tubuloside A had antioxidant activity and were concentration-dependent. Conclusion This study can provide evidence for the Q-Marker of CH, and provide a theoretical basis for quality control and evaluation of CH.
