基于DSS序列的皖产道地药材凤丹皮及其混伪品的鉴别

中文摘要:目的:建立一种快速、简便、准确的鉴别凤丹皮、白芍与赤芍的DNA特征序列(DSS)标记分子鉴定方法。方法:基于芍药属植物叶绿体基因组数据库,利用 IdenDSS 软件筛选出DSS序列组及其特异性引物对,经DNA提取、PCR扩增、测序、序列比对,进行凤丹皮目标DSS验证。结果: 通过5对候选DSS序列的验证,最终确定DSS-10:GTAACTTGCTATCCTCTTGCCTAGCAGGCAAAGATTTACT可用于风丹皮基原鉴定,并将上述序列上传至中药分子鉴定平台(www.herbsdna.com)标准数据库中,结合数据库BLAST功能,实现凤丹皮的快速、准确基原鉴定。结论:筛选的DNA特征序列标记可有效应用于凤丹皮的基原鉴定。

Study on the Original Identification of Adenophorae Radix Based on DNA Signature Sequence Tags
Abstract:Objective: A quick, convenient, and accurate DNA Signature Sequence Tags(DSS) molecular identification method was established to identify Moutan cortex,Paeoniae radix alba and Paeoniae radix rubra. Methods: DSS sequence group and its specific primers were screened using IdenDSS software based on the chloroplast genome sequences obtained from Chloroplast Genome Information Resource (CGIR). After DNA extraction, PCR amplification, sequencing, and sequence alignment verification, the Target DSS used to validate Moutan cortex were obtained. Results: The verification results of 5 candidate DSS sequences was ultimately determined that DSS-10: GTAACTTTGCTATCCTTGCCTAGCAGGCAAAGATTTACT can be used for the identification of Moutan cortex and its adulterants. The above sequence was uploaded to the Traditional Chinese Medicine Molecular Identification Platform (www.herbsdna. com), and combined with the BLAST function, rapid and accurate original identification of Moutan cortex was achieved. Conclusion: The screened DNA Signature Sequence Tags could be effectively applied to the original identification of Moutan cortex.
