中文摘要:目的 建立葶苈大枣泻肺汤物质基准的高效液相色谱法(HPLC)指纹图谱和6个指标成分的含量测定方法。方法 采用反相高效液相色谱梯度洗脱法,借助“中药色谱指纹图谱相似度评价系统”(2012A版)建立14批葶苈大枣泻肺汤的HPLC特征指纹图谱,同时测定槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷、芦丁、异槲皮苷、异鼠李素-3-O-β-D-葡萄糖苷、槲皮素和异鼠李素6个指标成分的含量。结果 葶苈大枣泻肺汤的HPLC特征指纹图谱共标定了27个共有峰,6个特征峰分别为5号峰(槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷)、18号峰(芦丁)、19号峰(异槲皮苷)、21号峰(异鼠李素-3-O-β-D-葡萄糖)、24号峰(槲皮素)和27号峰(异鼠李素)。对14批葶苈大枣泻肺汤进行含量测定,槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷、芦丁、异槲皮苷、异鼠李素-3-O-β-D-葡萄糖苷、槲皮素和异鼠李素质量浓度分别为111.33~118.71、9.16~9.36、16.51~17.26、12.23~12.62、3.10~3.36、3.51~3.62 μg·mL–1,方法精密度、稳定性、重复性均良好。结论 建立的HPLC指纹图谱和多指标成分含量测定方法稳定可靠,可为葶苈大枣泻肺汤的质量标准研究提供参考。

Evaluation of Tingli Dazao Xiefei Decoction Based on HPLC Fingerprint Technology
Abstract:Objective To establish the high-performance liquid chromatography (HPLC) fingerprint of Tingli Dazao Xiefei Decoction and a method for determination of six index components.Method The HPLC fingerprints of 14 batches of Tingli Dazao Xiefei Decoction were established by reverse-phase HPLC gradient elution method according to the similarity evaluation system for chromatographic fingerprint of Traditional Chinese Medicine (Version 2012A). At the same time, the contents of six index components (quercetin-3-O-β-D-glucose-7-O-β-D-gentiobiglycoside, rutin, isoquercitrin, isorhamnetin-3-O-β-D-glucoside, quercetin and isorhamnetin) were determined.Results A total of 27 common peaks were calibrated according to the HPLC characteristic fingerprints of Tingli Dazao Xiefei Decoction, and six chromatographic peaks were identified, including peak 5 (quercetin-3-O-β-D-glucose-7-O-β-D-gentiobiglycoside), peak 18 (rutin), peak 19 (isoquercitrin), peak 21 (isorhamnetin-3-O-β-D-glucose), peak 24 (quercetin), and peak 27 (isorhamnetin). The content determination for the 14 batches of Tengli Dazao Xiefei Decoction resulted in the following mass concentrations. The content of quercetin-3-O-β-D-glucose-7-O-β-D-gentiobiglycoside was 111.33-118.71 μg·mL–1, the content of rutin was 9.16-9.36 μg·mL–1, the content of isoquercitrin was 16.51-17.26 μg·mL–1, the content of isorhamnetin-3-O-β-D-glucoside was 12.23-12.62 μg·mL–1, and the content of quercetin was 3.10-3.36 μg·mL–1, and the content of isorhamnetin was 3.51-3.62 μg·mL–1. The established fingerprint and content determination method exhibited good precision, stability and reproducibility.Conclusion The established HPLC fingerprint profile and multi-index component quantification method are stable and reliable, which can provide a reference for the study on the material basis and quality standard of Tingli Dazao Xiefei Decoction.
