人参红皮病对人参皂苷含量的影响及其机制研究

中文摘要:目的:揭示人参红皮病对人参质量的影响及本质。方法:采集6月和8月的不同发病阶段人参韧皮部样品,分为AG(健康人参韧皮部组织)、BG(患病人参锈斑周围未变色组织)和CG组(患病人参锈斑组织),测定其O2?-、H2O2和MDA的含量及抗氧化酶SOD、CAT、POD和次生代谢相关酶HMGCR、FPS、SS、SE、DE、DS等的活性,并分析健康人参和患病人参的人参皂苷Rg1、Re、Rf、Rb1、Rc、Ro、Rb2、Rb3的含量。结果:不同月份取样的红皮病人参组织各指标的检测结果呈现相同的变化趋势,与AG组相比,BG组中ROS含量、抗氧化酶和次生代谢相关酶的活性均在达到最高,MDA含量在CG组中达到最高;与健康人参相比,6月份红皮病发病初期各人参皂苷的含量显著提高,8月份发病后期各人参皂苷的含量显著降低。结论:红皮病影响人参药材质量,在发病初期或胁迫程度较低的条件下皂苷含量较高,在严重胁迫条件或红皮病发病后期皂苷含量较低。

Investigation into the impact and mechanism of ginseng rusty symptoms on ginsenoside content
Abstract:Objective: To reveal the effect and essence of ginseng rusty root symptoms on ginseng quality. Methods: the phloem tissue of healthy ginseng (AG), phloem tissue around red-patches of rusty ginseng (BG), and red-patches tissue of rusty ginseng (CG) were collected,to detected the contents of O2?-, H2O2, and MDA, the activities of antioxidant enzymes (SOD, CAT and POD) and secondary metabolism-related enzymes (HMGCR, FPS, SS, SE, DE and DS). The contents of ginsenosides Rg1, Re, Rf, Rb1, Rc, Ro, Rb2, Rb3 in healthy ginseng and rusty ginseng were analyzed by HPLC. Result: The test results of the ginseng samples in different months showed the same change trend. Compared with the AG group, the contents of ROS, the activities of antioxidant enzymes and secondary metabolize-related enzymes in the BG group reached the highest, and the MDA content in the CG group reached the highest. Compared with healthy ginseng, the contents of ginsenosides significantly increased in the early stage of rusty ginseng in June and significantly decreased in the late stage in August. Conclusion: Ginseng rusty root symptoms can affect ginseng quality, with higher levels of saponins in the early stages of the disease or under less stressful conditions, and lower in the severe conditions or the later stages of the disease.
