三七片对急性软组织损伤大鼠的作用及机制研究

中文摘要:目的 探讨三七片对急性软组织损伤(acute soft tissue injury,ASTI)大鼠的治疗途径及作用靶点。方法 以重物击打致ASTI大鼠为研究对象,经三七片干预后,在不同时间节点通过对大鼠组织损伤评分,HE染色观察各组大鼠软组织病理形态变化并评分,以及血清中炎症因子IL-1β,TNF-α,PGE2和COX-2含量检测,考察三七片对ASTI大鼠损伤的治疗效果;检测大鼠软组织中COX-2和PGE2的蛋白表达水平及COX-2/EP2通路的表达量,探究三七片治疗ASTI大鼠的作用靶点及机制。结果 三七片能有效降低受损后大鼠血清中的IL-1β和TNF-α含量,抑制炎症反应,起到较好的抗炎效果。通过定量检测软组织损伤大鼠血清中COX-2、PGE2的含量,半定量检测炎症通路中COX-2、PGE2及EP2的表达,结果分析得知,三七片能降低大鼠血清中炎症因子COX-2含量,随着剂量的增加,在每个取材时间节点上,都具有一定的量效关系。结论 三七片通过降低ASTI模型大鼠血清中IL-1β、TNF-α、COX-2和PGE2的含量,抑制组织中COX-2,PGE2及EP2表达发挥抗炎作用。

Study on the effect and mechanism of Sanqi Tablet on rats with acute soft tissue injury
Abstract:Objective To investigate the therapeutic pathways and targets of Sanqi Tablets in rats with acute soft tissue injury (ASTI). Methods The therapeutic effect of Sanqi Tablets on ASTI rats was investigated after the intervention of Sanqi Tablets, and the pathological morphological changes of soft tissues in each group were observed and scored by tissue damage score and HE staining at different time nodes, and the contents of inflammatory factors IL-1β, TNF-α, PGE2 and COX-2 in serum were detected to investigate the therapeutic effect of Sanqi Tablets on ASTI rat injury. The expression of EP2 pathway was explored to explore the target and mechanism of Sanqi Tablets in the treatment of ASTI rats. Results Sanqi Tablets could effectively reduce the content of IL-1β and TNF-α in the serum of damaged rats, inhibit the inflammatory response, and have a good anti-inflammatory effect. By quantitatively detecting the contents of COX-2 and PGE2 in the serum of rats with soft tissue injury, and semi-quantitatively detecting the expressions of COX-2, PGE2 and EP2 in the inflammatory pathway, the results were analyzed and it was found that Sanqi Tablets could reduce the content of inflammatory factor COX-2 in the serum of rats. Conclusion Sanqi Tablets can exert anti-inflammatory effects by reducing the serum contents of IL-1β, TNF-α, COX-2 and PGE2 in ASTI model rats, and inhibiting the expression of COX-2, PGE2 and EP2 in tissues.
