防风多糖部分酸水解降解产物的免疫调节活性与结构研究

中文摘要:目的:基于防风多糖的部分酸水解降解产物探究防风多糖的免疫调节活性与结构特征。方法:选取不同浓度的三氟乙酸对防风多糖进行控制性降解,采用长春瑞滨(NVB)诱导的免疫低下斑马鱼模型评价防风多糖酸水解降解产物的免疫调节活性,并基于亲水作用色谱-蒸发光散射法(HILIC-ELSD)和超高效液相色谱-Q Exactive四极杆-静电场轨道阱串联质谱(UHPLC-Q-Orbitrap MS/MS)初步分析其降解产物。结果:实验结果表明,三氟乙酸能够有效裂解防风多糖的糖苷键,得到丰富的寡糖片段,并且不同条件下降解的防风多糖酸水解降解产物对NVB造成的免疫细胞损伤均具有治疗作用。酸水解条件越剧烈,降解生成的寡糖片段越丰富,提高斑马鱼免疫细胞密度的能力越显著。在寡糖片段最丰富的样品中共鉴定出42种酸水解降解产物,包括8种单糖、13种糖醛酸、21种寡糖(DP 2~5)。结论:通过酸水解方法降解结构复杂的多糖,测定相应的免疫调节活性,可以为多糖的构效关系研究提供依据。

Study on immunoregulatory activity and structure identification of partial acid hydrolysis degradation products of Saposhnikoviae Radix Polysaccharides
Abstract:Objective: To further study the immunoregulatory activity and structural characteristics of Saposhnikoviae Radix Polysaccharides (SP), controlling acid hydrolysis of SP was carried out using trifluoroacetic acid under different conditions. Methods: The partial acid hydrolysis degradation products of SP were used as the research object to evaluate the immunoregulatory activity by the zebrafish immunosuppression model, which used hydrophilic interaction chromatography-evaporative light scattering detector (HILIC-ELSD) for chemical characterization and ultra-high-performance liquid chromatography-Q Exactive quadrupole-electrostatic field-orbitrap tandem mass spectrometer (UHPLC-Q Orbitrap MS/MS) for preliminary structure identification. The results by indicated that the acid hydrolysis degradation products maintained good immune activity. With the more intense acid hydrolysis conditions, more abundant oligosaccharide fragments were generated by polysaccharide degradation, and the ability to improve the density of zebrafish immune cells were more significant. Results: Based on the acid hydrolysis condition that can obtain abundant oligosaccharide fragments, 42 types of acid hydrolysates were identified, including 8 monosaccharides, 13 uronic acids and 21 oligosaccharides (DP 2-5). Conclusion: The polysaccharides with the complex structure were acid hydrolyzed and determination of the corresponding changes in immunomodulatory activity, which may provide a foundation in the structure-activity relationship exploration of polysaccharides.
