Abstract:Objective To establish a mouse model of spleen deficiency and investigate the spleen-tonifying and digestion-promoting effects of water extract of Crepis phoenix Dunn.Methods The experimental animals were randomly divided into blank group, model group, positive control (Shenling Jianpiwei Granules of 10.3 g·kg-1) group, water extract groups of C. phoenix (1.0, 2.0, and 4.0 g·kg-1), and ethanol extract groups of C. phoenix (1.0, 2.0, and 4.0 g·kg-1). Except for the blank group, the mice in other groups were administered intragastrically with rhubarb combined with senna leaf to induce the mouse model of spleen deficiency syndrome. After successful modeling, mice in each group were given the corresponding sample by gavage at 20 mL·kg-1, once a day for seven consecutive days. The body weight, independent activity, body temperature, food utilization rate, spleen index, gastric emptying rate, small intestine propulsion rate, gastrointestinal hormone, and other indicators were used to evaluate the spleen-tonifying and digestion-promoting effect of C. phoenix.Results Compared with the blank group, the body weight increase, food utilization rate, body temperature, independent activity, serum motilin (MTL) level, serum gastrin (GAS) level, gastric emptying rate, and spleen index of the mice in the model group were decreased, while the serum vasoactive intestinal peptide (VIP) level and small intestine propulsion rate were increased, indicating that the mouse model of spleen deficiency syndrome was successfully established. Compared with the model group, the water extract of C. phoenix had a significant improvement in body weight increase, food utilization rate, body temperature, and spleen index in mice but slightly improved the independent activity, serum MTL level, serum GAS level, serum VIP level, gastric emptying rate, and small intestinal propulsion rate. The ethanol extract of C. phoenix had a significant improvement in body weight increase, food utilization rate, independent activity, serum MTL level, serum VIP level, gastric emptying rate, small intestinal propulsion rate, and spleen index in mice but slightly improved the body temperature and serum GAS level.Conclusion C. phoenix has obvious spleen-tonifying and digestion-promoting effects, and the spleen-tonifying effect of the ethanol extract is better than that of the water extract. The mechanism may be related to regulating gastrointestinal hormones and immunity. |
