中文摘要:目的:利用气相色谱-质谱联用技术(GC-MS)建立止咳桃花散的指纹图谱,并对其中挥发性成分进行含量测定,以评价止咳桃花散中挥发性成分的质量,为后续改良制备工艺提供参考。方法:使用HP-5MS毛细管色谱柱,氦气为载气,流速为1 mL·min-1,分流比为25:1,进样量为1 μL。检测器为质谱仪,检测器温度为250 ℃,进样口温度220 ℃。 柱温程序为:初始温度82 ℃保持20 min,10 ℃·min-1至205 ℃,保持7 min,再以20 ℃·min-1达到250 ℃。结果:确定止咳桃花散指纹图谱共有峰16个,核心特征峰11个,相似度均在0.99以上。其中,异龙脑、左旋龙脑、薄荷脑、麝香酮的浓度范围分别为:1.073~21.46、0.99~19.96、0.0869~1.390、0.021~0.538 μg·mL-1(r=0.9999),线性关系良好。结论:建立的止咳桃花散中挥发性成分含量测定方法简单易行、灵敏度高、结果稳定可靠,为中药质量评价提供参考。

Analysis of Volatile components in Zhike Taohua powder by Gas Chromatography-Mass Spectrometry
Abstract:Objective: Gas chromatography-mass spectrometry (GC-MS) was used to establish a fingerprint of Zhike Taohua powder and determine the content of volatile components to evaluate the quality of volatile components in Zhike Taohua powder, providing reference for subsequent improvement of preparation process. Method: The analysis was performed on a HP-5MS capillary column with helium as the carrier gas at a flow rate of 1 mL·min-1 with a 25:1 split ratio and a 1 μL injection volume. The detector was a mass spectrometer with a detector temperature of 250 ℃ and an inlet temperature of 220 ℃. The column temperature procedure was as follows: the initial temperature of 82 ℃ was held for 20 min, 10 ℃·min-1 to 205 ℃, held for 7 min, and then 20 ℃·min-1 to 250 ℃ without holding. Result: There were sixteen common peaks in the fingerprints of ten batches of samples with the similarities more than 0.99, and eleven peaks were identified. The concentration ranges of Isoborneol, L-borneol, Menthol, and Muscone were 1.073~21.46, 0.99~19.96, 0.0869~1.390, 0.021~0.338 μg·mL-1(r=0.9999), respectively, with good linear relationships. Conclusion: The stable,reliable and reproducible method can be used for the quality control of Zhike Taohua powder.
