中文摘要:目的:制备15批山西临汾不同采收时间的青翘标准汤剂,建立特征图谱,结合多元统计分析及多种质量指标评价其质量差异。方法:利用UPLC 建立青翘标准汤剂特征图谱,结合质谱识别特征峰,开展相似度计算、聚类分析、主成分分析;测定挥发油转移率、醇溶性浸出物、出膏率、连翘苷转移率、连翘脂苷A转移率,并开展优劣解距离法(technique for order preference by similarity to an ideal solution, TOPSIS)分析。结果:15个批次青翘标准汤剂通过特征图谱标出13个共有峰,并成功指认了13个成分,批间的相似度超过了0.900。15个批次样品可分为2类,其中采收前期的聚为一类,采收中末期的聚为一类,经主成分分析,青翘标准汤剂3个主成分累计贡献率达85.925%;TOPSIS分析表明,综合排名前5位的均为采收中末期样品。结论:建立的UPLC 特征图谱结合多元统计分析方法及多种质量指标可以评价不同采收时间山西临汾青翘标准汤剂的质量差异,并提供质控依据。

Analysis of Quality Differences of Premature Forsythia Fructus Standard Decoction in Linfen of Shanxi at Different Harvest Time
Abstract:Objective:To prepare 15 batches of standard decoction of Premature Forsythia Fructus in Linfen of Shanxi at different harvest time, establish characteristic chromatograms and evaluate the quality differences through multivariate statistical analysis and multiple quality indicators. Methods:The characteristic chromatogram of Premature Forsythia Fructus standard decoction was established by UPLC, and the characteristic peaks were identified by mass spectrometry, and the similarity calculation, cluster analysis and principal component analysis were carried out;the extraction rate of volatile oil, alcohol-soluble extract, extract rate, the transfer rates of forsythin and forsythoside A were determined, and TOPSIS method was analyzed .Results:13 common peaks were demarcated from 15 batches of standard decoction of Premature Forsythia Fructus and 13 components were successfully identified. The similarity between batches was over 0.900. The 15 batches of samples can be divided into two categories, among which the samples in the early stage of harvest were grouped into one category, and the samples in the middle and end of harvest were grouped into another category. By principal component analysis, the cumulative contribution rate of the three principal components of Premature Forsythia Fructus standard decoction reached 85.925%, and TOPSIS analysis showed that the top 5 samples were all collected in the middle and late stages. Conclusion: The UPLC characteristic chromatogram combined with multivariate statistical analysis and multiple quality indicators can evaluate the quality difference of Premature Forsythia Fructus standard decoction in Linfen of Shanxi at different harvest time, and provide the basis for quality control.
