中文摘要:目的 研究防风多糖SP-3的抗肿瘤活性及作用机制。方法 通过噻唑蓝(MTT)法、流式细胞术检测防风多糖SP-3对癌细胞增殖、细胞周期及凋亡的影响,采用秀丽隐杆线虫(以下简称线虫)模型研究防风多糖SP-3抗肿瘤活性机制。结果 防风多糖SP-3可以显著抑制胃癌AGS细胞、肝癌HepG2细胞的增殖,促进胃癌AGS细胞的凋亡并影响其细胞周期,且呈剂量依赖性;防风多糖SP-3对大鼠肉瘤(Ras)基因突变的线虫多阴门表型具有显著的抑制作用。结论 防风多糖SP-3具有良好的抗肿瘤效果,其作用机制可能与Ras/丝裂原活化蛋白激酶(MAPK)通路有关。

Antitumor Activity and Mechanism of Saposhnikovia divaricata Polysaccharide SP-3
Abstract:Objective To study the antitumor activity and mechanism of Saposhnikovia divaricata polysaccharide SP-3.Methods The effects of SP-3 on cancer cell proliferation, cell cycle, and apoptosis were detected using the MTT assay and flow cytometry. The antitumor activity mechanism of SP-3 was studied using the Caenorhabditis elegans model.Results SP-3 significantly inhibited the proliferation of gastric cancer AGS cells and hepatoma HepG2 cells, promoted apoptosis in gastric cancer AGS cells, and affected their cell cycle in a dose-dependent manner. Additionally, SP-3 exhibited a notable inhibitory effect on the multivulva phenotype in Ras gene mutant C. elegans.Conclusion SP-3 has a potent antitumor effect, and its mechanism may be related to the Ras/mitogen-activated protein kinase (MAPK) pathway.
