中文摘要:目的 探究海风藤分级多糖(KPSP)的抗阿尔茨海默病(AD)活性。方法 采用分级醇沉制备3种醇沉体积分数(40%、60%、80%)的KPSP(KPSP1~KPSP3),通过苯酚-硫酸法测定KPSP的糖含量;以CL4176秀丽隐杆线虫(以下简称线虫)为模型生物,观察KPSP对线虫寿命、瘫痪情况、应激能力及体内活性氧(ROS)自由基、超氧阴离子自由基和β淀粉样蛋白(Aβ)含量的影响。结果 KPSP1~KPSP3中糖质量分数分别为(57.45±0.27)%、(65.32±1.26)%、(70.26±3.53)%,结果显示最佳质量浓度下的KPSP1~KPSP3分别使线虫寿命延长了38.35%、42.02%和39.13%,在一定程度上可以减缓线虫的瘫痪速率,减少线虫体内ROS、超氧阴离子自由基和Aβ含量。结论 KPSP具有较好的抗氧化活性和抗AD活性。 |

Investigating Anti-Alzheimer's Disease Activity of Kadsura Pepper Stem Polysaccharide Based on Model Organism of Caenorhabditis Elegans |

Abstract:Objective To explore the anti-Alzheimer's disease (AD) activity of fractionated polysaccharides from Kadsura Pepper Stem (KPSP).Methods KPSP (KPSP1-KPSP3) was prepared with three ethanol precipitation concentrations (40%, 60%, and 80%) using graded ethanol precipitation. The sugar content of KPSP was determined using the phenol-sulfuric acid method. Caenorhabditis elegans CL4176 (hereafter referred to as nematodes) was used as a model organism to observe the effects of KPSP on nematode lifespan, paralysis, stress capacity, and contents of reactive oxygen species (ROS), superoxide anion radical and β-amyloid protein (Aβ) in nematodes.Results The sugar mass fractions in KPSP1, KPSP2 and KPSP3 were (57.45±0.27)%, (65.32±1.26)%, and (70.26±3.53)%, respectively. The results showed that KPSP at the optimal concentration prolonged the nematode lifespan by 38.35%, 42.02%, and 39.13%, respectively. To a certain extent, it slowed the paralysis rate of the nematodes and reduced the contents of ROS, superoxide anion radical, and Aβ in the nematodes.Conclusion KPSP exhibits good antioxidant activity and anti-Alzheimer's disease activity. |
