中文摘要:目的 研究太白美花草对肺炎幼鼠肠道菌群、短链脂肪酸(SCFA)代谢的影响,并探讨其潜在的作用机制。方法 以SD幼年大鼠为研究对象,利用16S rRNA高通量测序,检测幼年大鼠粪便中肠道菌群的变化情况;通过气相色谱-质谱法对粪便中的乙酸、丙酸、丁酸、异丁酸和异戊酸等SCFA含量进行测定。结果 太白美花草可提高幼年大鼠体质量(P<0.01),显著降低肺组织炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)的水平(P<0.05)和肺指数(P<0.01),改善肺损伤;可显著改善幼年大鼠肠道菌群紊乱,恢复肠道菌群的多样性,提升有益菌的相对丰度,提高粪便中乙酸、丙酸的含量(P<0.05);在门水平上,主要表现为厚壁菌门的相对丰度降低、拟杆菌门的相对丰度升高;在纲水平上,主要表现为拟杆菌纲的相对丰度上升,而梭状芽孢杆菌纲的相对丰度接近对照组;在目水平上,主要表现为拟杆菌目相对丰度升高,梭菌目相对丰度降低;在科水平上,主要表现为普雷沃菌科相对丰度显著提高,毛螺球菌科、瘤胃球菌科相对丰度显著降低;在属水平上,主要表现为普氏菌属-9、乳杆菌属、普雷沃菌属Ga6A1、普氏菌属-1、毛螺菌属NK4A136、普雷沃菌属UCG-001相对丰度有上升趋势,拟杆菌属、普拉梭菌、直肠真杆菌相对丰度有下降趋势。结论 太白美花草能通过调节肠道菌群和SCFA的含量改善肺炎幼鼠炎症因子的水平和肠道菌群多样性。 |

Regulatory Effects of Callianthemum taipaicum W. T. Wang on Inflammatory Factors and Intestinal Flora in Young Rats with Pneumonia |

Abstract:Objective To study the effects of Callianthemum taipaicum W. T. Wang on the intestinal flora and short-chain fatty acid (SCFA) metabolism in young rats with pneumonia and to explore its potential mechanisms of action.Methods Changes in intestinal flora in the feces of SD young rats were detected using 16S rRNA high-throughput sequencing. The content of SCFAs, including acetic acid, propionic acid, butyric acid, isobutyric acid, and isovaleric acid in feces, was determined by gas chromatography-mass spectrometry.Results C. taipaicum significantly increased the body weight of young rats (P<0.01), reduced the levels of inflammatory factors TNF-α and IL-8 in lung tissue (P<0.05), decreased the lung index (P<0.01), and improved lung injury. It also significantly alleviated disturbances in the intestinal flora of the young rats, restored the diversity of intestinal flora, increased the relative abundance of beneficial bacteria, and elevated the content of acetic acid and propionic acid in feces (P<0.05). At the phylum level, C. taipaicum primarily led to a decrease in the relative abundance of Firmicutes and an increase in the relative abundance of Bacteroidetes. At the class level, the relative abundance of Bacteroidia increased, while the relative abundance of Clostridia tended to be similar to that of the control group. At the order level, the relative abundance of Bacteroidales increased, while that of Clostridiales decreased. At the family level, the relative abundance of Prevotellaceae significantly increased, while that of Lachnospiraceae and Ruminococcaceae significantly decreased. At the genus level, there was an increasing trend in the relative abundance of Prevotella_9, Lactobacillus, Prevotellaceae_Ga6A1_group, Prevotella_1, Spirillum NK4A136 group, and Prevotella_UCG-001, while the relative abundance of Bacteroides, Faecalibacterium, and [Eubacterium]_rectale_group decreased.Conclusion C. taipaicum can improve the levels of inflammatory factors and the diversity of intestinal flora in young rats with pneumonia by regulating the composition of intestinal flora and the content of SCFAs. |
