中文摘要:目的 以不同国家(地区)穿心莲为研究对象,通过指纹图谱、化学模式识别和多成分含量测定的方法分析不同产地穿心莲质量。方法 建立26批穿心莲药材指纹图谱,进行相似度评价分析,采用化学模式识别方法对26批穿心莲药材进行主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA),寻找各产地穿心莲的差异性成分,并进行含量测定。结果 穿心莲指纹图谱有15个共有峰,除S2、S4和S15外,其余23批相似度均大于0.8;PCA将26批穿心莲药材按产地划分为2类,OPLS-DA进一步筛选出3个差异性标志物,分别是穿心莲内酯、14-去氧穿心莲内酯和脱水穿心莲内酯;含量测定结果显示,国外采集的穿心莲中二萜内酯成分普遍高于国内样品。结论 建立的指纹图谱方法结合化学模式识别能有效分析不同产地穿心莲药材质量的差异性,且国内外不同产地穿心莲中二萜内酯含量具有明显差异,为穿心莲药材质量及种植提供参考。 |

Quality Evaluation of Andrographis paniculata from Different Countries and Regions |

Abstract:Objective The quality of Andrographis paniculata from different countries and regions was analyzed by fingerprint, chemical pattern recognition, and multi-component content determination.Methods The similarity of 26 batches of A. paniculata was evaluated and analyzed with the fingerprints established in this study. Chemical pattern recognition was employed for principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) to find the differential components of A. paniculata from different producing areas, and content determination was also performed.Results There were 15 common peaks in the fingerprints of A. paniculata. Except for the three batches of samples S2, S4, and S15, the similarity of the other 23 batches was greater than 0.8. According to PCA, the 26 batches of A. paniculata were divided into two categories. Three differentiation biomarkers were determined by OPLS-DA, namely andrographolide, 14-deoxyandrographolide, and dehydroandrographolide. The content of diterpene lactone in A. paniculata from other countries was generally higher than that from China.Conclusion The established fingerprint method combined with chemical pattern recognition can effectively analyze the quality difference of A. paniculata from different producing areas, and the diterpene lactone content of A. paniculata from China and other countries is significantly different. This study provides a reference for the quality improvement and planting of A. paniculata. |
