中文摘要:目的:基于高通量靶点筛选-靶标关联-临床应用的中药复方靶标研究方法,精准阐释疏风解毒胶囊(SFJDC)的作用靶标及发挥抗H1N1病毒感染新机制。方法:进行动物实验,评估SFJDC对H1N1病毒入侵小鼠肺部后的影响;采用高通量靶点筛选技术CETSA-MS,对SFJDC在BEAS-2B细胞中的整体作用靶点进行高通量筛选;利用IPA(Ingenuity Pathway Analysis)数据库对SFJDC作用靶点与作用靶标及疾病特异性差异表达蛋白质进行关联研究。结果:本研究发现SFJDC可以降低病毒入侵小鼠肺组织中病毒RNA水平、肺指数以及IL-1、IL-6和TNF-α等炎症因子的表达,具有较好的抗病毒功效;通过CETSA-MS筛选获得SFJDC直接作用靶点150个、间接作用靶点66个;通过IPA数据库分析,其中42个直接作用靶点和18个间接作用靶点与抗病毒功效相关,能够在病毒复制和感染等多个环节发挥干预治疗作用。并且SFJDC可能参与调控干扰素信号通路、MAPK/NF-κB信号通路、JAK1-STAT1信号通路、Toll样受体信号通路和内吞作用通路等多条通路,发挥抗病毒功效。结论:本研究运用高通量靶点筛选-靶标关联-临床应用的中药复方靶标研究方法,基于SFJDC的抗病毒功效,从靶标角度阐明SFJDC发挥抗病毒功效的作用机理。 |

Abstract:Objective: Based on the high-throughput target screening-target association-clinical application target research methodology of traditional Chinese medicine, the target and the new mechanism of anti-H1N1 virus infection of Shufeng Jiedu Capsules (SFJDC) were precisely elucidated. Methods: Animal experiments were conducted to assess the effects of SFJDC on H1N1 virus invasion in the lungs of mice; CETSA-MS, a high-throughput target screening technology, was used to conduct high-throughput screening of the overall targets of SFJDC in BEAS-2B cells; IPA (Ingenuity Pathway Analysis) database was used to conduct correlation studies between SFJDC's targets of action and downstream targets and disease-specific differentially expressed proteins. Using IPA (Ingenuity Pathway Analysis) database, the association between the target of action and downstream antiviral targets and disease-specific differentially expressed proteins was studied. Results: In this study, it was found that SFJDC could reduce the level of viral RNA, lung index, and the expression of inflammatory factors such as IL-1, IL-6 and TNF-α in the lung tissue of virus-invaded mice, and it had good antiviral efficacy; 150 direct targets and 66 indirect targets of SFJDC were obtained through the screening of CETSA-MS; and 42 direct targets and 18 indirect targets were related to antiviral efficacy in the analysis of the IPA database. Through IPA database analysis, 42 direct targets and 18 indirect targets are related to antiviral efficacy, which can play a therapeutic role in various aspects of viral replication and infection. Moreover, SFJDC may participate in the regulation of interferon signalling pathway, MAPK/NF-κB signalling pathway, JAK1-STAT1 signalling pathway, Toll-like receptor signalling pathway and endocytosis pathway to exert antiviral efficacy. Conclusion: In this study, we applied the high-throughput target screening-target association-clinical application target research methodology of traditional Chinese medicine to elucidate the mechanism of the antiviral effect of SFJDC from the target perspective, based on the antiviral effect of SFJDC. |
