中文摘要:目的 以马钱子为例,提出一种优化中药炮制方法的半定向转化策略,建立更高效、可标准化的现代炮制方法,获得药效更优、毒性更低的马钱子炮制品。方法 搭建通氧加热炮制装置,以加热温度、加热时间、加样量为考察因素,以士的宁、马钱子碱、士的宁氮氧化物、马钱子碱氮氧化物含量的总评归一值(OD)为监测指标,采用Box-Behnken响应面法优化炮制工艺。结果 马钱子的最优通氧炮制工艺为加样量4.8 g、加热温度180 ℃、加热时间1 h。经3次验证,所得平均OD为93.15(RSD=1.68%),与预测OD(97.20)的相对误差为4.17%。结论 提出了一种半定向转化策略,并成功建立了马钱子通氧炮制方法,有效提高了士的宁氮氧化物和马钱子碱氮氧化物在马钱子炮制品中的含量,可为中药炮制现代化研究提供新的思路。

Simulated Processing Methods of Strychni Semen Based on Semi-directional Transformation Strategy
Abstract:Objective With Strychni Semen as an example, a semi-directional transformation strategy was proposed to optimize the processing method of traditional Chinese medicine (TCM), and a more efficient and standardized modern processing method was established to obtain Strychni Semen products with better effects and less toxicity.Methods An oxygenation heating processing device was established. Heating temperature, heating time, and sample size were taken as factors for investigation, and the overall desirability (OD) values of strychnine, brucine, strychnine N-oxide, and brucine N-oxide content were taken as monitoring indexes. The Box-Behnken response surface method was used to optimize the processing process.Results The optimal processing conditions of Strychni Semen by oxygenation were as follows: sample size of 4.8 g, heating temperature of 180 ℃, and heating time of 1 h. After three experiments, the average OD score is 93.15 (RSD=1.68%), and the relative error between the average OD score and the predicted OD score (97.20) is 4.17%.Conclusion In this study, a semi-directional transformation strategy was proposed, and an oxygenation processing method of Strychni Semen was successfully established, which effectively improved the content of strychnine N-oxide and brucine N-oxide in the processed Strychni Semen products and could provide a new idea for the research on the modernization of TCM processing.
